The AS400 does allow you to turn off/on any card in the buss (ie: for install/maintenance) without turning off the AS400. I've installed SCSI cards this way while the machine is running. No big deal. The thing to watch out for, as noted below, is to make sure you can just reset that one card and not have to reset the entire IOP which might (probably will) have other cards on it in which case it might be ok and it might not depending on the cards sharing the IOP.
