Jonathan, What are you trying to use to access the 400? 5250 Terminal emulation (which one?) Since you're directing traffic to port 80 on the 400 I assume you are just using http, iSeries Access for the web perhaps? Are you sure only one port is needed? Client Access for instance needs more than just the tn5250 port.
