To do it precisely the way you stated, you'd need the 5250 emulator to have support for your file dialog, etc. Then, it'd have to have the capability to upload the file to the i, which would involve server-side code as well. That's probably not going to happen.

You can get very close, however, by integrating other technologies besides green screen. Your 5250 emulator could invoke a PC program, and the PC program could present the file dialog, and could have code to handle the upload.

Or, probably the most obvious solution, the F-key could invoke a web browser on the PC. The browser could present a file upload dialog, and upload the file. Unlike a 5250 emulator, a web browser has this capability built-in. I've done this many times.
