We use sharepoint but not really as a front end to anything. We have links that take people to iSeries web sites and we played around with using one of the sharepoint standard web parts to run a web service and bring back some data. That was just a simple RPG CGI app that read some data, and sent back some HTML to the sharepoint web part that displayed the HTML to the user. I have not investigated 2010 yet as we just installed it but in 2007 typical Microsoft would not let you connect to any database other than MS SQL. There you could shows lists of data from an MS SQL Table fairly easily but if your data was in any other database you could only access it through a web service.
