If you monitor the VPN logs you will know.

Plus if they have a soft phone I believe you can monitor that as well.

Then there's video surveillance, but if you have to go that far you have a bigger issue :-)

In our environment, some work from home, connected via VPN, but then remote desktop to work computer.
In this scenario, you will not be able to distinguish.
