Once you get the failure again, the profile is probably locked and needs to be unlocked again. Navigator has the locked list somewhere in the Netserver configuration. I don't have access to it to walk you through the steps, but I think it was under Network and Servers perhaps. When you see Netserver, right click on it to find something about locked accounts.
On your test user - does that user exist as a System i user? It can't very well disable a user who doesn't exist. There is also "public" or "guest" mapping to allow unauthenticated users access depending on your settings.
With the way Windows sends the password repeatedly, I don't think you'll get anywhere without the passwords synchronized. After changing the Windows password, I always make users log out of Windows and back in to reset the cached credentials, or it continues to fail. That's with browsing through Explorer, but I suspect mapping a drive from a prompt will yield the same results.

I did the CHGUSERPRF for her but still got the original error. I then tried changing her password from a green screen but still got the original error. I gone through System/i Navigator but haven't found anything that would indicate she's locked out. Network and AS/400 passwords don't match, but then it worked for my test user-id which didn't match.

Check that the username and passwords match between Windows and System i.
Check DSPLOG MSGID(CPIB682) to see if the user was disabled for file
sharing. Enable by using CHGUSRPRF USERNAME with no other parameters or
dig through Navigator to find and unlock users.
